In vitro: standard culture of tumor cell lines and of different primary cell types
SERVICE DESCRIPTION:
The cell cultivation includes primary cultures of animal cells as well as established cell lines. Members of this laboratory optimized standard culture of cancer cell lines (melanoma, colon carcinoma) as well as in vitro models for processes involved in tumor development (inflammation hypoxia) by using co-culture of cancer cells and macrophages under different experimental conditions. The infrastructure for cell culturing is provided by Molecular Biology Centre belonging to Institute for Interdisciplinary Research in Bio‑Nano‑Sciences, Babes‑Bolyai University, where our group is also affiliated.

Animal facility and handling of laboratory animals
SERVICE DESCRIPTION:
The animal facility houses rodents (mice and rats) and sparrows kept under standard conditions (food and water available ad libitum, and a 12 h light/dark cycle). Rats are used for animal behavior experiments and for testing various therapeutic plant extracts. Sparrows are used for ecophysiological studies (stress-dependent immunological resistance and tolerance against specific parasites (e.g. coccidians)) . Our group also optimized two in vivo murine tumor models (B16.F10 melanoma and C26 colon carcinoma) via subcutaneous tumor cell inoculation. Experiments are performed according to the national regulations and are approved by the Babes-Bolyai University ethical committee.

Biostatistical data analysis
SERVICE DESCRIPTION:
ENVIRONMENTAL: Standardized data are used for correlation analysis of environmental variables and canonical correspondence analysis using Microsoft Excel 2013/XLSTAT-ADA (Addinsoft, Inc., Brooklyn, NY, USA) with the significance level set at p<0.05. Cluster analysis is performed using PAST software (Hammer et al. 2001). BIOMEDICAL: Data from different experiments are usually reported as mean ± SD. Statistical comparisons of the overall effects of different treatments on tumors are evaluated by one-way ANOVA with Dunnett’s post-test for multiple comparisons. The differences between the effects of different treatments on angiogenic/inflammatory factor production are analyzed by two-way ANOVA with Bonferroni correction for multiple comparisons using GraphPad Prism version 6 for Windows, GraphPad Software (San Diego, CA). A value of P<0.05 is considered significant.

Microbial (algal, fungal, bacterial, archaeal) cell isolation, cultivation, manipulation and maintenance
SERVICE DESCRIPTION:
Culture-dependent methods are employed for isolation of microbial and fungal strains by means of selective and rich-medium followed by genotypic and phenotypic characterization. Genotypic analysis consists of total DNA extraction followed by PCR amplification of SSU rRNA-gene sequence and subsequent identification by bioinformatics. Phenotypic description of isolated strains comprise growth characteristics, substrate utilization, Gram-staining, antibiotic susceptibility, cell and colony morphology, etc.

Nucleic acid separation and manipulation (extraction, PCR amplification, electrophoretic separation and purification)
SERVICE DESCRIPTION:
Extraction, separation and purification of DNA is performed using appropriate extraction/purification kits according to manufacturer's instruction followed by PCR amplification of targeted gene sequence by thermocycler device. Quality check (QC) of nucleic acids is performed spectrophotometrically and by agarose-gel horizontal electrophoresis (Bio-Rad systems) and subsequent verification by UV trans-illuminator or UVP imaging system.

Bioinformatic analyses of next-generation sequencing data
SERVICE DESCRIPTION:
The metabarcoding analyses will be performed using QIIME, mothur and obitools software with versatile data analysis pipelines. The metagenomic and metatranscriptomic sequences will be trimmed using sickle software and de novo assembled using the IDBA-UD and IDBA-MT assemblers. For the predictions of protein coding gene we will use the assembled contigs (obtained following assembly) and Prodigal software in metagenomic mode. For the tRNAs prediction we use tRNAscan-SE software and for the gene annotations the NCBI NR, COG and TIGRfam databases. Supplementary, the full and partial genomes will be annotated using the RAST server. The rRNA genes present in metagenomes will be identified using ssu-align and meta_rna software and classified using Greengenes database. For the genome reconstruction we will use only the manually curated contigs and we will bin them into MAGs (Metagenome-assembled genomes) using the software MetaCluster and MBBC. The MAGs will be checked for genome completeness and contamination using CheckM software and will be further classified using PhyloPhlAn. For metabolic reconstruction we use the software MetaPhlAn and HUMAnN2. For high quality representations of taxonomic and phylogenetic trees we implement GraPhlAn software.

Molecular cloning, microbial community fingerprinting and qPCR estimation of microbial cell counts
SERVICE DESCRIPTION:
By using plasmid vectors we can construct, screen and analyse DNA and cDNA clone libraries. Additionally, we offer means to quickly profile and interpret the diversity of microbial communities by using PCR-based techniques such as: T-RFLP, ARISA and DGGE. We offer relative and absolute gene quantification by using the real-time PCR technique with DNA-binding dyes.

PAR sensors measure the flux of quanta (unit, μmol m-2 s-1) in the photosynthetically relevant wavelength range, that is, between 400 and 700 nm. Pyranometers detect the power of radiation (unit, W m-2) mostly in a wider spectral range so that solar radiation arriving at the Earth’s surface can be measured. When power of radiation is measured, a flux of short-wavelength (e.g. blue) photon results in a higher signal than the same flux of photons with longer wavelengths (e.g. red) simply because the former flux carries more energy than the latter.
