SERVICE DESCRIPTION The research service addresses the investigation of the the interaction of ionizing radiation with human normal cells using the expression profile of stress genes profile. The research service encompasses the following investigations: 1. Cultivation and multiplication of human SC monocytes 2. Investigations on cell viability (the MTS reduction test, in conjunction with the LDH release test), apoptosis and necrosis (flow cytometry with Annexin V-propidium iodide) and proliferation (flow cytometry with CFDASE) 3. Preparation of cells for RNA extraction 4. Evaluation by qRT-PCR of the expression profile of 84 stress genes and 5 housekeeping genes (Stress and toxicity pathway finder, Qiagen) 5. Gene expression data processing and selection of candidate stress genes 6. Individual validation of selected genes by qRT-PCR 7. Data interpretation.

DESCRIPTION:
CytoViva’s technology combines patented, enhanced darkfield optical microscopy with high resolution hyperspectral imaging in the visible near-infrared (VNIR 400nm-1,000nm) wavelength range. These integrated technologies were specifically designed to enable optical observation and spectral characterization of label free nano-materials as they interact with biological and other materials-based matrixes. No fluorescent labeling or other sample prep is required to optically observe or spectrally characterize nano-materials in these complex environments. At 100x magnification, a hyperspectral image may contain as many as 700,000 pixels as small as 128 nm each. This spectral data is recorded at approximately 2 nm of spectral resolution in the VNIR range, enabling minute spectral differences to be measured from pixel to pixel within the image. This can be accomplished without any special staining or tagging of the target material, as is often required with traditional microscopy techniques. When using the CytoViva Dual Mode Fluorescence system, researchers can also observe the interactions between fluorescently labeled nanoparticles (NPs) or bacteria and live (u)nlabeled cells, along with biological processes within the cells.
