Luminescence dating electron spin resonance dating methods have multiple applications in archaeology, geology, palaeoclimatology and geo-archaeology. These dating methods allow the direct determination of the moment of deposition in a variety of sedimentation environments with the upper age limit of 1Ma depending on the method and material used. Radioactive dating using Pb-210 can be used for dating recent sediments, that were deposited during the last 200 years. Information stored in natural sedimentary archives is used in a wide range of applications, such as observing changes in pedological erosion processes or monitoring atmospheric pollution with heavy metals, organic pollutants, radioactive emissions. Studies on natural and artificial radioactivity by gamma spectrometry Among the identification and quantification methods of radioactive pollutants, the gamma spectrometry stands out as being a method characterised by a large applicability, in both radioactive products which it can be used for, and sample types (soil, sediment, water, air, organic matter etc.), with a very good precision, a simple sample preparation and a relatively short measurement time. The Gradient Palm–Cycler™ from Corbett is a user-friendly gradient thermal cycler; it is available in a 96-well or 384-well format. Individual 0.2 ml tubes or strip tubes can be used. The temperature range of operation is from 4ºC to 99ºC with 8 Peltier devices engaged in fast heating and cooling. This allows for the enrichment of the correct product over any non-specific product. This option avoids the lengthy programming, as is often necessary with other thermal cyclers. Similarly, a long-range parameter allows cycle lengths to be changed in designated increments after a particular cycle number is reached.

DNA sequencing experiments determine the order of the bases in a DNA sample. Fluorescently labeled dyes are attached to ACGT extension products in DNA sequencing reactions. Dye labels are incorporated using either 5´-dye labeled primers or 3´-dye labeled dideoxynucleotide terminators. Polymerases such as AmpliTaq® FS are used for primer extension. The sequencing reaction sample tubes are placed in a tray in the instrument’s autosampler. The autosampler successively brings each sample into contact with the cathode electrode and one end of a glass capillary filled with polymer. An anode electrode at the other end of the capillary is immersed in buffer. A portion of the sample enters the capillary as current flows from the cathode to the anode. This is called electrokinetic injection. The end of the capillary near the cathode is then placed in buffer. Current is applied again to continue electrophoresis. When the nucleotides reach a detector window in the capillary coating, a laser excites the fluorescent dye labels. Emitted fluorescence from the dyes is collected by a CCD camera. The software interprets the result, calling the bases from the fluorescence intensity at each data point.

Each row of eight samples (sample set), containing labeled DNA fragments, is automatically denatured and then separated by capillary electrophoresis. The replaceable medium (separation gel) is automatically replaced in the eight capillaries after each separation. The separation gel supply is an easily replaced cartridge with a capacity sufficient for a two full microplates. Detection is by laser-induced fluorescence in four spectral channels. The four-channel raw data sets generated by each of the eight capillaries are automatically processed to produce high quality base sequences or fragment lists after separation. Raw and analyzed data are stored in a database and may also be exported in file formats compatible with common analysis applications.
