The core facility of IRASM Center (http://www.nipne.ro/facilities/facilities/irasm.php) is the Multipurpose Irradiator, which is a gamma irradiator with Cobalt-60 radiation sources. Radiation sterilization
SERVICE DESCRIPTION:
Radiation sterilization is a well established and standardized method for sterilization of single use medical supplies and pharmaceutical products. We are using Cobalt-60 gamma radiation in an industrial irradiator which can sterilize up to 10 m3 of materials at the same time. We are cooperating with partners from industry in RDI projects or by contract, for the development of new products and for product qualification to radiation sterilization, including the sterilization validation.

Irradiation for testing of materials
SERVICE DESCRIPTION:
Radiation resistance of materials is of great interest for various industries such as medical devices, nuclear power plants or aerospace. Among many types of radiation currently used for material testing, gamma is the most common because of its high availability in research or industrial irradiators, well standardized dosimetry and high reproducibility of the irradiation experiments. IRASM Department of IFIN-HH acquired a long experience in setting up gamma irradiation for different materials. It operates two Co-60 gamma irradiators: a research, self-contained irradiator, and an industrial, versatile, panoramic irradiator. Irradiation setups are available for a dose range from 0,01 kGy to over 1000kGy and a dose rate range from 0.1kGy/h to more than 10kGy/h.

Irradiation treatment for conservation of cultural heritage artifacts
SERVICE DESCRIPTION:
Many cultural heritage items are threatened by biological attacks, the organic constituents (cellulose, collagen or proteins) being food for microorganisms and insects. Improper storage conditions lead to massive biologic attacks in museums, archives or libraries, which are difficult to stop or eliminate because of the large quantities of items involved. Compared to other techniques, gamma radiation treatment has a major advantage: the biocide effect is ensured by the radiation`s high penetration power. At a molecular level, the photons or electrons break the molecular bonds initiating further chemical reactions. The scission of the DNA double coil is the major cause for the biocide effect and the basis of the applications related to “killing” the pests (bacteria, fungi or insects). The physical measure which best describes the gamma radiation effects is the absorbed dose – Gy unit. In order to reduce the risk of degrading the treated item, establishing the correct irradiation dose for a particular class of materials is of major importance.

Microbiological testing
SERVICE DESCRIPTION:
IRASM Microbiological laboratory offers a comprehensive range of microbiology services complying with GMP requirements for sterile and non-sterile product testing and facility monitoring. The laboratory is fully equipped to analyze all the range of pharmaceutical products (sterile or non-sterile) and medical devices. The clean rooms (HEPA-filtered) ensure contamination-free testing. The bioburden test is used to estimate present total number of viable micro-organisms and the absence of specific pathogens. The sterility test is used to detect the presence of viable microorganisms. A critical step is the preparatory testing which validates the product with the method used. This ensures the product has no inhibitory effect on the recovery of microorganisms from the product.

Study of cultural heritage artifacts by vibrational spectroscopy, thermal analysis and chromatography methods
SERVICE DESCRIPTION:
FTIR and Raman vibrational spectroscopic methods are complementary techniques used for identifying and characterizing materials and organic/inorganic substances by applying nondestructive or minimal destructive tests. Infrared Spectroscopy provides more information on the organic component of the painting layer, and Raman spectroscopy is a nondestructive and non-contact analysis method usedmainly for the study of pigments. Thermal analysis tests (TG/DSC) are used fot the determination of oxidation and thermal decomposition temperatures. Analysed matrices: fossil resins, painting materials, paper, wood, leather, parchment, wool, textiles etc. Chromatographic tests are used for: fatty acids profiling from microbial, biological and organic matrices; identifying volatile organic compounds by thermal desorption from the materials. Elemental and isotopic analysis by Induced Coupled Plasma Mass Spectrometry (ICP-MS) allows the elemental and isotopic structural characterisation of materials by minimal destructive analysis: approx. 70 elements and 130 isotopes detection limit on ppb level (ng/g).

Determination of radiation resistance of microbial isolates

PCR-RFLP Genotyping of Filamentous Fungi
SERVICE DESCRIPTION:
PCR-RFLP (Polymerase Chain reaction Restriction Fragment Length Polymorphism) is a method of molecular typing, proven useful for rapid characterization of filamentous fungi species. The method is especially intended for characterization of a strain, especially in regulated areas like biotechnology. We use different fungal primers, targeting either the Internal Transcribed Spacer (ITS) region, small (18S), large (28S) subunit of ribosomal DNA (rDNA) region or coding genes, to generate a PCR amplification product. After purification, this product is digested with different restriction enzymes, and the fragments are separated and analyzed by gel-electrophoresis. The pattern is compared between similar strains. Through extensive studies, a polymorphism may be found which is specific for a particular strain. The work for developing this service was supported by a grant of the Romanian Ministry of Education and Research, CCCDI- UEFISCDI, project number PN-III-P2-2.1-PTE-2019-0373 / 36PTE/2020, within PNCDI III.

Multipurpose Gamma Irradiator

The gamma irradiator is of class IV (source storage in a pool, automatic transport system), product overload and tote-box type. The goods transport system integrates an external conveyor with goods loading and unloading stations placed in the storage and maze and an internal conveyor placed into the irradiation room. The irradiation begins when the sources are lifted from the water pool in the air, in the middle of a metallic structure supporting internal conveyor. Totes are pneumatic moved around the sources, in 52 positions. In each position an incremental dose is delivered to the incidence of total presetting dose. At the irradiation end, the source comes back in the pool and the totes caring irradiated goods evacuates the irradiation room using the external conveyor. The irradiator was built with IAEA support (IAEA ROM/8/011).
