Proangiogenic activity of biomaterials conditioned as powders, discs, films or nanomaterials is evaluated using human mesenchimal stem cell cultures (hMSC). hMSC are cultivated with sterile biomaterials or with cell growing media incubated with said materials. After 7 or 14 days, growing media or cell lysate are subjected to multiplex analysis and results are read with Luminex 200™ platform. Data statistical analysis gives the proangiogenic activity of a given biomaterial.

The ProteinChip surface-enhanced laser desorption/ionization (SELDI) system’s high resolving ability combines the separation power of two techniques – chromatography and high-sensitivity mass spectrometry for protein profiling, biomarker discovery, and immunoassay applications. This allows large numbers of proteins and peptides to be detected and profiled, so you can rapidly generate a list of candidate disease biomarkers. The ProteinChip SELDI system’s high throughput allows you to easily acquire sufficient data for statistical significance, making the biomarker discovery process more efficient. Differential expression profiling — the comparison of protein expression levels across different disease or treatment states — helps identify proteins involved in disease and provides a direct method for monitoring disease progression and the therapeutic effects of drugs. The ProteinChip SELDI system integrates a ProteinChip SELDI reader with data analysis software to analyze proteins selectively captured on ProteinChip arrays. ProteinChip arrays selectively bind and retain whole classes of proteins from complex samples. The mass profile of the bound proteins is read by the ProteinChip SELDI reader, creating high-resolution protein maps. The unique surface chemistries of ProteinChip arrays allow on-chip profiling — the selective capture and analysis of peptides and proteins on a single platform.

DESCRIPTION: 2D electrophoresis is one of the most powerful protein separation techniques available. It has the ability to resolve complex mixtures of thousands of proteins simultaneously in a single gel. In the first dimension, proteins are separated by isoelectric point (pI), while in the second dimension, they are separated by molecular weight. Standardization of this technique has simplified the process and improved gel-to-gel reproducibility. Depending on the experiment, requirements for resolution, throughput, and speed may vary. For example, when sample preparation conditions are being developed, speed is more important than resolution or throughput. Whereas resolution is more important for analyzing complicated proteomes, throughput is important when many samples must be run. We have all components of the 2D process: isoelectric focusing system, mini/large capacity gel electrophoresis, system imaging and analysis.
